激光诱变选育高产PUFAs真菌菌株的研究

摘要
采用PIV400型Nd:YAG激光器,对少见报道的产PUFAs丝状真菌菌株Monoblepharis polymorpha Cornu进行激光诱变育种.在激光照射剂量:波长532 nm,功率50 mW,时间10 min下,孢子致死率为75%~80%.突变株中ω-3系列PUFAs正突变率大于80%,ω-3系列突变株FPAm59传代10代以上性能稳定,在土豆基质上摇瓶培养6 d后,生物量达到8 g/L,产油率53.4%,GLA 343.1 mg/L、AA 1209.0 mg/L、EPA 125.1 mg/L、DHA 119.4 mg/L; 分别比对照菌株提高了2.5倍、9.7倍、18.0倍、6.6倍、10.3倍、3.8倍.结果表明,激光诱变育种是获得PUFAs高产菌株尤其是生产ω-3系列PUFAs菌株的有效方法.
Abstract
In order to obtain mutant fungi strain with highly content PUFAs. The fungi spore in the plate were radiated at wavelength 532 nm for 10 minutes using Nd:YAG laser,the lethality ratio of spores is approximately 75%~80%.The mutation rate of series ω-3 is above 80%.the mutation strain FPAm59 can retain its stability during more than 10 passages.After six days culture,the biomass of FPAm59 is up to 8 g/L and 2.5 times of the control strains.The lipid component of the biomass is 53.4% and (9.7) times of control strains.The quantity of the GLA,AA,EPA,and DHA in the strain lipid is 343.1mg/L, 1 209.0 mg/L,125.1mg/L,119.4mg/L and 18.0 times,6.6 times,10.3 times,and 3.8 times of control strains lipid.Therefore,laser mutation to fungi is an effective method to obtain the high quantity of PUFAs especially to the ω-3 series.
