摘要
基于含硫核苷类化合物的光敏性和核苷类化合物对肿瘤细胞有特殊的亲和力以及核苷类化合物的抗肿瘤活性, 设计合成了4-硫胸苷, 并通过 NMR、UV、MS对其结构进行了表征。利用荧光光谱考察了4-硫胸苷与人血清白蛋白(HSA)的相互作用。实验结果表明: 4-硫胸苷对人血清白蛋白(HSA)体系的猝灭是由于他们相互作用形成复合物而导致的静态猝灭。通过计算得出其结合常数分别为3.46×104, 3.91×104, 4.31×104 L·mol-1, 并测定了不同温度下的热力学参数。结果表明, 4-硫胸苷与人血清白蛋白之间的主要作用力类型为疏水作用力。另外, 通过圆二色光谱考察了4-硫胸苷对人血清白蛋白二级结构的影响, 结果表明, 4-硫胸苷对人血清白蛋白的二级结构基本没有影响。
Abstract
Based on photosensitivity and a special affinity on tumor cells and anti-tumor activity for thionucleoside compounds, 4-thiothymidine was designed and synthesized. The structure was characterized by NMR, UV, MS. The interactions between 4-thiothymidine and human serum albumin (HSA) were also investigated by the fluorescence spectroscopy. The results show that 4-thiothymidine on HSA has a static fluorescence quenching, the binding constants of 4-thiothymidine and HSA are 3.46×104, 3.91×104, 4.31×104 L·mol-1, respectively. The determination of thermodynamic parameters at different temperatures for 4-thiothymidine and HSA was investigated, the results show that the main forces of both interactions have typical hydrophobic interaction from thermodynamic data to determination. In addition, the effect of 4-thiothyminidine on the secondary structure of HSA was investigated by circular dichroism, the results show that 4-thiothyminidine has no effect on the secondary structure of HSA.
