近红外光谱技术在快速测定连翘提取物中连翘苷含量的应用

摘要
本文主要对近红外光谱技术在快速测定连翘提取物中连翘苷含量的应用进行了研究。利用近红外漫反射光谱法采集样品的近红外光谱, 以HPLC分析值作为参考值, 采用偏最小二乘法(PLS)建立连翘苷含量的定量校正模型, 并用未知样品对该模型进行验证。所建模型的相关系数(R2)、校正均方差(RMSEC)和内部交叉验证均方差(RMSECV)分别为0.9745、0.117和0.2392;经外部验证, 预测相关系数(r2)和预测均方差(RMSEP)分别为0.9788和0.0776。结果表明该方法操作简便, 无污染, 结果准确可靠, 可用于连翘提取物中连翘苷含量的快速测定。
Abstract
This paper concentrates on the determination of phillyrin in extractum forsythiae siccus by near-infrared spectroscopy technique. Through collecting near-infrared spectra by near-infrared diffuse reflectance spectroscopy technique,with HPLC analysis values as reference,the quantitative calibration medol was built by partical least squares(PLS), and validated with the unknow samples.The correlation coefficients(R2),the root-mean-square error of calibration(RMSEC) and the root-mean-square error of cross-validation(RMSECV) of the quantitative calibration model for phillyrin was 0.9745,0.117 and 0.2392; the correlation coefficients of prediction(r2)and the root-mean-square error of prediction(RMSEP) were 0.9788 and 0.0776. The results indicated that the method is simple,non-polluted and accurate,and could be applied for the fast determination of phillyrin in extractum forsythiae siccus.
