激光波长对荧光激发谱分布影响研究

摘要
激光器是DNA测序仪中的重要部件,寻求合适波长的激光器可有效提高DNA测序仪性能。分析了DNA检测所用的四种荧光染料FAM、JOE、TAMRA和ROX的激发谱和发射谱,设计实验技术方案利用488 nm、505 nm和515 nm三种不同波长的激光器分别对其进行激发。在相同的积分时间和相同的功率下,488 nm激光器对于荧光染料ROX的激发强度太低, 515 nm激光器对于荧光染料FAM的激发强度太低,而505 nm激光器对于四种荧光染料的激发强度均比较强。实验结果表明在三种不同波长中,505 nm激光器对四种荧光染料的激发效果最佳,可以替代目前大多数DNA测序仪中的氩离子激光器。
Abstract
Laser is an important component in DNA sequencing instrument. It is necessary to seek lasers with appropriate wavelength to effectively improve the performance of DNA sequencing instrument. Excitation spectra and emission spectra of the fluorescent dye FAM, JOE, TAMRA and ROX used in DNA detection are analyzed. They are excited by the 488 nm, 505 nm and 515 nm lasers respectively in the design experiment technology solution. At the same integration time and power, the excited intensity of the 488 nm laser is low for ROX, the 515 nm laser is low for FAM, and the 505 nm laser is strong for all four luorescent dyes. The experimental results show that in the three different wavelengths, the excited effect of 505 nm laser on four kinds of fluorescent dyes is best and can replace the Ar+ laser widely used in DNA sequencing instrument.
