N+注入Lactobacillus bulgarius诱变选育高产ACE抑制剂的研究
A Study on High-yielding Strains of ACE Inhibitory Peptides through the N+ Implanting into Lactobacillus bulgarius
摘要
采用N+注入技术对实验室保存的4株保加利亚杆菌L601、L602、L603、L606进行诱变处理, 旨在选育出高产血管紧张素转换酶(angiotensin converting enzyme, ACE)抑制剂菌株。结果表明, 在离子注入能量为20 keV, 剂量为3.0×0.5×1015 ions/cm2条件下, 诱变效果较好。从正突变菌株中反复筛选, 获得1株对ACE抑制活性较高的菌株M602, 其ACE抑制率为82.49 %。经过培养条件优化, 其对ACE抑制率进一步提高为85.12 %, 经连续传代实验, 其性状遗传稳定。
Abstract
The aim of this paper was to obtain high-yielding strains of ACE inhibitory peptides through the N+ implanting into four strains of Lactobacillus bulgarius (L601, L602, L603 and L606). The results showed that the best mutation condition were that energy is 20 keV, and dose is 3.0×0.5×1015 ions/cm2. The strains M602 with stronger ACE inhibitory activity was obtained from the mutant strains and its ACE inhibitory rate was 82.49 %.After improving the culture conditions, the ACE inhibitory rate increased to 85.12 %, and it can be inherited stably.
