啤酒花节间外植体再生体系的建立

摘要
在国内首次建立了两个啤酒花主栽品种(麒麟丰禄, 青岛大花)的节间外植体再生体系。实验表明: 啤酒花无菌苗的节间为适于不定芽再生的外植体; 适宜的再生培养基为MS基本培养基+B5维生素+葡萄糖20 g/L, 附加IAA 0.25 mg/L+TDZ 2 mg/L(麒麟丰禄)或IAA 0.125 mg/L+TDZ 1.5 mg/L(青岛大花), 以7 g/L琼脂固化, pH5.8; 不定芽再生频率可达57 %(麒麟丰禄)或16 %(青岛大花)。啤酒花再生体系的建立, 为利用植物基因工程技术培育抗病虫害啤酒花新品系奠定了基础。
Abstract
The regeneration systems from internode explants of two main cultivars of hops (Humulus lupulus L.) are established for the first time in China. The experiments showed that: the shoot internodes from in vitro cultures of hops were the suitable explant type for shoot regeneration; the optimal regeneration medium was MS basal medium combined with B5 vitamins and glucose 20 g/L, supplemented with IAA 0.25 mg/L and TDZ 2 mg/L (for Qilinfenglu), or IAA 0.125 mg/L and TDZ 1.5 mg/L (for Qingdaodahua), and solidated with agar 7 g/L, pH5. 8; and the shoot regeneration frequency reached 57 % for Qilinfenglu and 16 % for Qingdaodahua in the separate optimal medium. The establishment of hop regeneration systems lays the foundation for the development of new hop lines with enhanced disease- and pest-resistance through plant genetic engineering techniques.
