308 nm准分子光对白癜风豚鼠模型Treg和Th17细胞的影响

摘要
目的: 通过白癜风豚鼠模型, 观察308 nm准分子光的疗效及对Treg、Thl7细胞的影响, 探讨紫外光疗机制中的免疫学通路。方法: 通过文件检索使用化学脱色法制备白癜风豚鼠模型并筛选造模成功的小鼠作为实验对象, 将小鼠分为实验组和对照组, 另选取相同数量的健康小鼠作为空白组, 对实验组进行308 nm准分子光的干预治疗, 造模成功的对照组以及健康小鼠的空白组均不予以处理, 10周后分别从肉眼复色程度、组织病理及免疫组化结果中的改变、mRNA以及蛋白表达的水平观察IL-17细胞和Treg细胞在三组之间的实验过程中的变化趋势。结果: 308准分子光对白癜风豚鼠模型达到治疗效果, 经过10周治疗后, 5只豚鼠达到完全复色。在组织病理中, 白癜风模型组豚鼠皮损中表皮黑素细胞减少, 伴随真皮淋巴细胞浸润。治疗后, 从组织学水平观察到, 黑素细胞的增多, 以及淋巴细胞浸润减少。免疫组织化学染色对皮损中IL-17标记可见, 造模成功组中真皮层可见阳性结果的出现, 此模型可用于本课题的研究。通过对皮损中IL-17及Foxp3的mRNA及蛋白表达水平定量检测发现, 模型组IL-17表达升高, Foxp3表达降低, 治疗后IL-17表达水平降低, Foxp3表达水平升高。结论: 308 nm准分子光疗可以通过降低豚鼠白癜风皮损中Th17细胞水平, 升高Treg细胞水平调节皮损中Th17和Treg细胞的平衡紊乱。
Abstract
Objective: Through the guinea pig model of vitiligo, observe the influence of 308 nm excimer light on Treg, Thl7 cells, explore the mechanism of ultraviolet phototherapy in immunological pathways.Methods: Using chemical decolorization to establish vitiligo guinea pig model and screening out the successful mice as experimental object.Then mices can be divided into the experimental group and control group, meanwhile selecting the same number of healthy mice as blank group.The experimental group of 308 nm excimer light intervention treatment, building the success of the control group and the blank group of healthy mice are not dealt with.After 10 weeks,from multicolor degree to the naked eye, histopathological and immunohistochemical results in the change, mRNA and protein expression level of observation of IL-17 cells and Treg cells between the three groups of the change trend in the process of experiment.Results: After 10 weeks after the treatment, 5 guinea pigs sucessfully recolored.In the lesions of guinea pig skin detect melanin cells decreased, with leather lymphocytic infiltrates in histopathological test.After treatment, from histological level observed, melanocyte increases, and lymphocyte infiltration reduction.Immunohistochemical staining was detected in the lesions of the IL-17 tags can be seen, dermis in building successful group shows positive results, this model can be used in the study of this subject.Based on the detected in IL-17 and quantitative test of mRNA and protein expression of Foxp3 levels found, model group, IL-17 higher expression, Foxp3 expression is reduced, IL-17 expression levels reduced after treatment,and Foxp3 expression levels upgrade.Conclusion: The 308 nm excimer phototherapy can reduce the Th17 cell level in guinea pig of vitiligo, elevated level of Treg cells to regulate balance disorders of Th17 and Treg cells.
