植物叶片微观结构对荧光测量的影响分析

摘要
为了更好地研究叶绿素荧光特性和分析植物生长状况, 提出了从植物叶片微观结构的特点出发分析影响叶绿素荧光测量的一种研究方法。采用波长为460nm的LED作为光源激发叶绿素荧光, 经滤光片后, 通过CCD分别获取宏观叶绿素荧光和微观叶绿素荧光, 对荧光图像进行预处理并获得叶绿素荧光比率Rfd均值; 在显微镜下用CMOS获取叶片的显微图像, 测量上表皮厚度、栅栏组织厚度和叶片厚度等微观结构几何参数; 统计分析了叶片微观结构对宏观荧光测量的影响情况。结果表明, 对于像植物这种复杂的活体被测对象, 测量时应该考虑植物叶片微观结构对植物叶绿素荧光测量的影响。
Abstract
In order to better study and analyze the chlorophyll fluorescence characteristics and the plant growth conditions, a research method was proposed to analyze the influence of chlorophyll fluorescence measurements from plant leaf microstructure. The excitation source for exciting chlorophyll fluorescence was 460nm LED. After the filter, CCD received the macroscopic chlorophyll fluorescence and microscopic chlorophyll fluorescence respectively. Then fluorescence image was processed and the average value of chlorophyll fluorescence ratio Rfd was obtained. The microstructure geometric parameters such as epidermis thickness, palisade thickness and leaf thickness were measured after receiving the leaf microscopic image by CMOS under microscope. Finally, the influence of plant leaf microstructure on the macroscopic fluorescence measurement was analyzed statistically. The results show that the effect of plant leaf microstructure on chlorophyll fluorescence measurement must be considered in the measurement of complex living objects such as plants.
